Unlock new characters and modes.

Secret Stages:
Brinstar Depths - Play 50 Vs. mode matches to unlock this stage.
Fourside - Play 100 verse mode matches to unlock this stage.
Big Blue - Play 150 verse mode matches to unlock this stage.
Poke' floats - Play 200 verse mode matches to unlock this stage.
Mushroom Kingdom (subcon) - Get the Birdo trophy(off of Super Mario Brothers 2) to unlock this stage.
Flat Zone - Beat Classic mode with Mr. Game&Watch to unlock this stage.
BattleField - Beat All-Star mode with any Character to unlock this stage.
Final Destination - Beat all 51 Events to unlock this stage.
Dream Land (64) - Beat break the Targets with all 25 characters to unlock this stage.
Yoshi's Island (64) - Get over 1400 ft in the home run contest to unlock this stage. The best way I've found is to use Yoshi and jump real low and use his down+A move in the air (he kicks a bunch of times). Doing this once should do around 33% damage to the sandbag, so do it a few more times, then grab the bat (which is to the left of you when you start by the way) and smash (tap right and hit A) the sandbag.
Kongo Jungle(64) - Finish the 15 minute melee with any character. I've found that DK is best (with his down+B move), but you can experiment.

Unlock Jigglypuff:
Beat the game on any difficulty level to unlock Jigglypuff; or beat the game in Classic or Adventure mode.

Unlock Luigi:
In adventure mode, at the first level you play the one with the koopas and goombas and beat the level in xx:x2:xx. In other words, beat the level where the tome in the fourth level slot is two. Instead of fighting Mario and Peach, Mario will be replaced by Luigi. Beat both of them in at least 45 seconds. Beat the game as you normally would and you'll fight Luigi again; and if you will have him where Pikachu normally was, don't worry Pikachu is in another place in the bottom of character select.

Other Method: Beat the first level in the Adventure mode with a 2 in its seconds (i.e., 6:32).

Unlock Mewtwo: play for 20 hours.
Unlock Falco: Beat 100-man melee.
Unlock Mr. Game and Watch: Beat classic mode with all 24 characters.
Unlock Marth: Beat classic mode with the first 14 characters.
Unlock Roy: Beat classic mode with Marth.
Unlock Pichu: Beat event match 37.
Unlock Dr. Mario: Beat classic or adventure mode with Mario.
Unlock Young Link: Beat classic mode with 10 characters; 2 of them must be Zelda and Link.
Unlock Ganadorf: Beat event match 17.
